READ ME File For 'Keable_thesis_dataset_basement_membrane_analysis_of_cerebrovascular_cells' Dataset DOI: 10.5258/SOTON/D1469 ReadMe Author: ABBY KEABLE, University of Southampton This dataset supports the thesis entitled: An in vitro model of intramural periarterial drainage. Significance for Alzheimer's diease. AWARDED BY: University of Southampton DATE OF AWARD: 2020 DESCRIPTION OF THE DATA Data was obtained by imaging in vitro cell cultures immunostained for proteins of interest using a confocal microscope. Images were exported to ImageJ and either threshold analysis or RGB measure tools were used to quantify the amount of staining present. Three images per experiment were captured and analysed and an average value per experiment was calculated. Raw values and averages were recorded in microsoft Excel. MTS absorbance was measured at 490nm with a Fluorstar optima plate reader and results were exported direct to Microsoft Excel with blank values subtracted. Raw data is presented in a spreadsheet and is best viewed with Microsoft Excel This dataset contains: Threshold analysis of basement membrane immunostaining in ApoE astrocytes after 72 hours in culture and at 100% confluence Fluorescence intensity values obtained from ImageJ RGB measure for basement membrane immunostaining in cerebrovascular cells at 72 hours Manual cell count opf astrocytes and their processes obtained from scanning electron micrographs of ApoE astrocytes Fluorescence intensity values obtained from ImageJ RGB measure for basement membrane immunostaining in cerebrovascular smooth muscle cells exposed to hypoxia and amyloid beta Fluorescence intensity values obtained from ImageJ RGB measure for basement membrane immunostaining in cerebrovascular smooth muscle cells exposed to millifluidic flow of amyloid beta MTS absorbance values for smooth muscle cell cultures exposed to hypoxia and amyloid beta at various timepoints Fluorescence intensity values obtained from ImageJ RGB measure for adrenergic receptor immunostaining in contractile brain cells at 72 hours Related projects/Funders: BBSRC Licence: CC BY https://creativecommons.org/licenses/by/4.0/ Date that the file was created: July, 2020
