Abstract
Intensive research into the leukotrienes as inflammatory mediators in asthma has tended to overshadow the intriguing evidence suggesting their involvement in cardiovascular disease. Cysteinyl-leukotrienes (Cys-LTs) fulfil many of Dale’s postulates for a pathophysiological contribution to angina, myocardial infarction and pulmonary hypertension. The development of safe and specific Cys-LT receptor antagonists now provides an opportunity to delineate their role in these conditions definitively. This paper will outline the data from animal and human studies in vitro and in vivo that suggest that Cys-LT contribute in particular to coronary artery disease (CAD) and that point towards new therapeutic roles for anti-LT drugs.
