Many immunological abnormalities are described in the peripheral blood of patients with Hodgkin's disease (HD) and relatively few observations were published describing the functional activities of lymphocytes in the lymphoid tissues of this disease. In this study the ability of the whole mononuclear cells extracted mainly from HD spleens to mediate NK activity against K562 and ADCC against CRBC and Chang cells were analysed and compared to that of a control group. Lysis of K562 was significantly increased in 15 uninvolved Hodgkin's spleens in comparison with 20 controls. The cytotoxicity was highest in the early and localized stages of the disease, and patients with B symptoms expressed significantly lower NK activity by splenic mononuclear cells than a-symptomatic (Stage A) patients. The ability of HD patients spleen mononuclear cells to lyse antibody coated CRBC (an effector capacity reduced by passage through Sephadex G-10 column) was not increased significantly neither was that against antibody coated Chang cells. In the view of the previous studies of lymphocyte subpopulation changes and lymphocyte function in the lymphoid tissues of this disease; the results of NK activity in this study provide further evidence of immunological activation of the spleen in HD prior to its involvement by the disease process, and may support the concept of ecotaxopathy in the pathogenesis of the disease.
