Boyd, Mark Thomas (1988) Detection of retroviral indicators in myeloproliferative diseases. University of Southampton, Doctoral Thesis.
Abstract
Previous work both in the U.S.A and Switzerland has suggested the possibility of a retroviral aetiology for the myeloproliferative disorders, polycythaemia vera and essential thrombocythaemia. The evidence for this was equivocal and the work presented here is an investigation of these diseases, with the intention of clarifying this situation. Evidence is presented for the existence of reverse transcriptase activity being present in the platelets of patients with these disorders. Electron microscopy of sucrose gradient fractions in which this enzyme activity has been detected, displayed particles which are morphologically indistinguishable from retroviral particles prepared in the same way. Northern blot analysis failed to detect the presence of RNA with any homology to a variety of retroviral probes, though this cannot exclude the possibility that a retrovirus may be present. (DX86648)
