A study of lanthanide chelates for use in radio-immunoscintigraphy and therapy
This work reviews in detail the current technology developing in the area of radio-immunoscintigraphy and therapy, the present use in tumour directed monoclonal antibodies and criteria for choice of radio-nuclide. It indicates that the lanthanide elements provide a wide range of suitable radio-isotopes for in vivo use and the related area of lanthanide co-ordination chemistry is examined. The thermodynamic stability of various lanthanide complexes is considered, relating conditional stability constant, ligand competition and metal speciation to pH, indicating the effectiveness of amino-polycarboxylate and amino-polyphosphonate ligands. The kinetics of dissociation of various lanthanide complexes with amino-polycarboxylate ligands are investigated and their lability on incubation in serum studied. Production routes to various bifunctional chelating ligands containing these functionalities are considered together with steps leading to their synthesis.
