Two images of a sample 300 are obtained. Each image indicates the intensity of Rayleigh scattered light 203 i) for incident light 201 of a different wavelength, or ii) for incident light 201 of a different polarisation state, or iii) of scattered light 203 of a different polarisation state. Alternatively, each image indicates the intensity of Raman scattered light 203 i) of a different wavelength, or ii) for incident light 201 of a different polarisation state, or iii) for incident light 201 of a different polarisation state. Chemical and/or material specific information of the sample 300 is determined based on the change in intensity of the scattered light 203 in dependence on the change in wavelength and/or polarisation of the incident and/or scattered light. The determination step may involve a look-up table, direct calculation, classification via e.g. principal component analysis or a neural network. Light sheet microscopy may be used.
