READ ME File For 'Dataset for: The Regulation of Daily Activity and Feeding Rhythms of the Economically Impactful Lepidopteran Pest Plutella xylostella' Dataset DOI: 10.5258/SOTON/D2567 ReadMe Author: Connor J Tyler, University of Southampton ORCID ID 0000-0002-9849-4074 This dataset supports the thesis entitled AWARDED BY: University of Southampton DATE OF AWARD: 2023 DESCRIPTION OF THE DATA Data files contain Excel files with data used for specific figures and analyses. Raw files contain original data collection files from TriKinetics recordings, imagery and Applied Biosystems qPCR machine outputs. TriKinetics locomotor activity recording hardware and software was used to produce txt files of activity counts. This was analysed using ClockLab Analysis Version 6 to produce data stored in the data files Excel files and locomotor activity figures in thesis. Larval feeding imagery was collected using a Bushnell 24mp trophy cam HD producing JPG image files. These were analysed in ImageJ also producing JPG files. Data from ImageJ analysis stored in the data files Excel files. qPCR data was collected using an Applied Biosystems StepOnePlus Real-Time PCR system. The associated StepOne Software v2.3 was used to operate and collect data from the produced eds files. Collected data stored in the data files Excel files. Other collected data is stored directly into Excel files with labelled Excel tabs showing associated figures and or tables. Stored data was used in Graph Pad Prism and R to produce analyses of activity, feeding and rhythms, including for the production of associated figures. Further details of collection methods and analyses are detailed in the Methods and Materials section (Chapter 2) of my thesis. This dataset contains: Data files zip - containing Excel data files for: Adult ClockLab data (Locomotor data), Larval data (feeding and weight change data), Lit review (data for keyword search in Web of Science), qPCR Adult (qPCR of circadian genes in adult samples), qPCR Larvae (qPCR of circadian and sulfatase genes in larval samples). Adult locomotor raw files zip - containing text files of locomotor count data for use in ClockLab or other related software. The above zip file contains: Cage exp (population cage experiment data) split into DD cage (constant dark experiment data) and LD cage (12/12 day/night cycle experiment data), Light cycles (varying light cycle experiment data) split into DD and LL (constant dark and constant light experiments) and changing light/dark phase length (12/12 = 12 hours of light followed by 12 hours of dark), Semi field (June and April UK environmental mimic conditions) split into April and June (specific UK month mimicking conditions detailed in thesis), Temperature matching (re-do of light cycle experiments in 12/12 and D/D to match conflicting results from a published paper). IP is split into 2 circadian genes, Per and Tim, with biological repeats 1-6 detailed and 3 sulfatase genes, Sulf2, Sulf3 and Sulf23 (Sulf2+3), with biological repeats 1-6 detailed, with both then split into technical replicates (various file naming). OP is split into 2 circadian genes, Per and Tim, with biological repeats 1-6 detailed and 2 sulfatase genes, Sulf2 and Sulf3, with biological repeats 1-6 detailed, technical repeats are detailed within eds files. A. thaliana imagery raw files zip - containing raw and analysed (using ImageJ) image files of larval feeding on A. thaliana. The above zip file contains: Col-0 wildtype A. thaliana (A.t WT), clock mutant A. thaliana (CCA1 OX and TM) and jasmonic acid mutant A. thaliana (AOS, COI1-16 and OPR3) feeding experiment as well as feeding preference between clock mutants and WT (Feeding between clock mutants). These files are split into 1212 (12/12 day/night cycle) and LL (constant light) experiments. B. rapa imagery raw files zip - containing raw and analysed (using ImageJ) image files of larval feeding on B. rapa.
