Scotopic central field focal-ERGs were conducted by projecting a 1.5mm (approximately 7.7-disc diameters) circular LED white light stimulus diameter, at a flash strength of 6.8log cd-s/m2 on to the central retina white LED light for 1ms. Two sweeps per eye were carried out with an interval time of 120 s, from which average a and b-wave amplitudes and implicit times were calculated. Leica Envisu R2200 VHR SDOIS Mouse Imaging System was used to obtain 1.4 mm volumetric scans (consisting of 100 B-scans and 1000 A-scans per B-scan), centred on the optic disc. Optomotor responses to horizontally drifting, vertically oriented gratings and vice versa were observed and scored using the OptoMotry system (Cerebral Mechanics, Inc.; Lethbridge, Canada). Methods for processing the data: Automatic segmentation of the individual retinal layers was carried out using InVivoVue 3.3 Diver software. Software- or Instrument-specific information needed to interpret the data, including software and hardware version numbers: None Standards and calibration information, if appropriate: N/A Environmental/experimental conditions: For electroretinography and optical coherence tomography examinations, mice were anesthetized with a mixture of ketamine (100 mg/kg, Chanelle) and xylazine (10 mg/kg, Bayer) in 0.1 mL in saline. Pupils were dilated using tropicamide 1% eye drops (Bausch & Lomb) and phenylephrine hydrochloride 2.5% eye drops (Bausch & Lomb) and Corneas were lubricated with a coupling gel (Clinitas gel, Altacor). Artificial tears (Systane Ultra, Alcon) were used throughout the procedure to maintain corneal moisture and clarity. For electroretinography examinations, mice were dark adapted overnight and subsequently handled under dim red-light illumination. Animals were positioned on a heated pad to maintain constant body temperature. Subdermal electrodes were used as the ground electrode inserted in the lumbar region, and as the reference electrode placed between the eyes. For OptoMotry assessments, awake and freely moving mice were place on a platform located in the centre of a chamber surrounded by four computer monitors with a camera positioned overhead.All mice were habituated to the space by placing them on the platform and showing them the gratings for a few minutes in each direction. The assessments took place during the second half of the light phase and each assessment lasted around 10 to 15 min per mouse.
