READ ME File For Dataset in support of the Southampton doctoral thesis 'The role of extracellular vesicle miRNA released in response to influenza A virus' Dataset DOI: 10.5258/SOTON/D2682 ReadMe Author: Laura Reid, ORCID ID 0000-0002-9994-4639 This dataset supports the thesis entitled The role of extracellular vesicle miRNA released in response to influenza A virus AWARDED BY: Univeristy of Southampton DATE OF AWARD: 2023 DESCRIPTION OF THE DATA This dataset contains: miRNA sequencing was completed for extracellular vesicles isolated from BCi either uninfected (n=5) or IAV infected at 3,600,000 IU/ml for 24 hours (n=5). EVs were isolated by filtration and size exclusion chromatography from the apical surface wash of air-liquid interface differentiated BCi. The EV miRNA profile was then sequenced, and reads were mapped to miRBase. Sequenced by Qiagen on a NextSeq (Illumina Inc.) sequencing instrument according to the manufacturer instructions. The following sequencing parameters were used: • Read type - Single-end read • Number of sequencing cycles (read length) - 75 nucleotides • Average number of reads - 12 million reads/sample Read processing was carried out by Qiagen using CLC Genomics Server 21.0.4. Qiagen mapped the reads to human miRBase version 22 using the workflow “QIAseq miRNA Quantification” of CLC Genomics Server with standard parameters.
