Abstract
This dataset includes raw and processed files from affinity purification mass spectrometry (AP-MS) experiments and live-cell imaging to characterise the interactomes and cellular dynamics of KIAA1671 and KIAA1143 in U2OS cells. It comprises the following components: AP-MS Proteomics Data KIAA1143_and_KIAA1671_proteomics.zip: This archive contains raw proteomics data (e.g., spectral counts, peptide information, and protein IDs) obtained from U2OS wild-type (WT) cells overexpressing GFP-tagged KIAA1671 or KIAA1143. Each AP-MS run was performed following GFP-affinity purification to identify potential protein interactors under interphase conditions. KIAA1671_CDK_AS_proteomics.zip: This file contains raw proteomics data from U2OS CDK1^AS (analog-sensitive) cell lines expressing KIAA1671-GFP. These experiments were designed to compare mitotic interactors of KIAA1671 under chemically synchronised CDK1^AS conditions. Contaminant Filtering and Interaction Scoring CRAPOME Excel.xlsx: This spreadsheet includes the output from the Contaminant Repository for Affinity Purification (CRAPome). It provides filtered AP-MS data with common contaminants removed and ranks high-confidence interactors based on the SAINT (Significance Analysis of INTeractome) scoring algorithm. This enables prioritisation of specific protein interactions for further validation. Live-Cell Imaging Data KIAA1671-GFP_H2B Cells movies.zip: A collection of live-cell imaging movies of U2OS cells expressing KIAA1671-GFP and H2B to visualise nuclear and centrosomal dynamics across the cell cycle. KIAA1143-GFP_movie.zip: A time-lapse movie capturing the localisation and dynamic behaviour of GFP-tagged KIAA1143 in live U2OS cells.
